Project Title: Synthesis of new TLR4 modulators from ligand- and structure-based rational design

Objectives: Formation of an ESRs with a solid knowledge of both synthetic and medicinal chemistry, training in industry. On the basis of structure-activity data of cationic and anionic amphiphiles previously developed by UNIMIB as TLR4 modulators, new molecules will be designed and synthesized with higher avidity for CD14 and MD-2 receptors and improved pharmacokinetic properties. These molecules will be based on glycolipid structures and on molecular simplification of Lipid A structure. The ESR will be trained on modern synthetic approaches, from the retrosynthetic analysis to stereochemistry control, and the use of homogeneous catalysts and enzymes.

Project Title: Synthetic variants of TLR4 modulators from plants and bacteria

Objectives: Formation of an ESRs with a solid knowledge of both synthetic and medicinal chemistry, training in industry. Some plant secondary metabolites (such as curcumin, taxanes, several phenols) and Lipid A variants from bacteria are active in modulating the TLR4 signal. Compound from plants present interesting solubility and distribution properties that are not associated to the TLR4 modulators mimicking Lipid A so far developed. The ESR will be involved in a project aimed at mimicking the natural compounds in synthetic molecules in the perspective to find new TLR4 modulators with improved distribution and bioavailability properties.
