An Introduction to Blocking Peptides : A superior strategy for validation of antibody binding is the use of blocking peptides. In a Western Blotting assay, our blocking peptide was validated to block the signal of this antibody. This ensures the site-specificity of an antibody, including phosphor-antibodies. In the case of the phosphor- antibodies, the antibodies can and will only recognize the phosphorylated target protein at the corresponding site, not the non-phosphorylated target protein or target protein phosphorylated at a different site.

Background : Serine/threonine-protein kinase component of the chromosomal passenger complex (CPC), a complex that acts as a key reg µLator of mitosis. The CPC complex has essential functions at the centromere in ensuring correct chromosome alignment and segregation and is required for chromatin-induced microtub µLe stabilization and spindle assembly. Plays also a role in meiosis and more partic µLarly in spermatogenesis. Has redundant cell µLar functions with AURKB and can rescue an AURKB knockdown. Like AURKB, AURKC phosphorylates histone H3 at \'Ser-10\' and \'Ser-28\'. AURKC phosphorylates the CPC complex subunits BIRC5/survivin and INCENP leading to increased AURKC activity. Phosphorylates TACC1 another protein involved in cell division, at \'Ser-228\'.

Involved in the bipolar attachment of spindle microtub µLes to kinetochores and is a key reg µLator for the onset of cytokinesis during mitosis. Required for central/midzone spindle assembly and cleavage furrow formation. Key component of the cytokinesis checkpoint, a process required to delay abscission to prevent both premature resolution of intercell µLar chromosome bridges and accum µLation of DNA damage : phosphorylates CHMP4C, leading to retain abscission-competent VPS4 (VPS4A and/or VPS4B) at the midbody ring until abscission checkpoint signaling is terminated at late cytokinesis (PubMed : 22422861 PubMed : 24814515). AURKB phosphorylates the CPC complex subunits BIRC5/survivin, CDCA8/borealin and INCENP. Phosphorylation of INCENP leads to increased AURKB activity. Other known AURKB substrates involved in centromeric functions and mitosis are CENPA, DES/desmin, GPAF, KIF2C, NSUN2, RACGAP1 SEPT1 VIM/vimentin, GSG2/Haspin, and histone H3. A positive feedback loop involving GSG2 and AURKB contributes to localization of CPC to centromeres. Phosphorylation of VIM controls vimentin filament segregation in cytokinetic process, whereas histone H3 is phosphorylated at \'Ser-10\' and \'Ser-28\' during mitosis (H3S10ph and H3S28ph, respectively). A positive feedback between GSG2 and AURKB contributes to CPC localization. AURKB is also required for kinetochore localization of BUB1 and SGOL1. Phosphorylation of p53/TP53 negatively reg µLates its transcriptional activity. Key reg µLator of active promoters in resting B- and T-lymphocytes : acts by mediating phosphorylation of H3S28ph at active promoters in resting B-cells, inhibiting RNF2/RING1B-mediated ubiquitination of histone H2A and enhancing binding and activity of the USP16 deubiquitinase at transcribed genes.

Background : AKIP (AURKA-interacting protein), also known as AURKAIP1 (aurora kinase A interacting protein 1) or AIP, is a 199 amino acid protein that localizes to the nucleus and is ubiquitously expressed, with highest levels present in testis, heart and skeletal muscle. Interacting specifically with ARK-1 (aurora kinase 1), AKIP functions to induce the proteasomal-dependent degradation of ARK-1 thereby acting as a negative reg µLator of ARK-1 activity. AKIP is encoded by a gene which maps to human chromosome 1 which spans 260 million base pairs, contains over 3,000 genes and comprises nearly 8% of the human genome. Chromosome 1 houses a large number of disease-associated genes, including those that are involved in familial adenomatous polyposis, Stickler syndrome, Parkinson’s disease, Gaucher disease, schizophrenia and Usher syndrome. Aberrations in chromosome 1 are found in a variety of cancers, including head and neck cancer, malignant melanoma and m µLtiple myeloma.

Background : Mitotic serine/threonine kinases that contributes to the reg µLation of cell cycle progression. Associates with the centrosome and the spindle microtub µLes during mitosis and plays a critical role in various mitotic events including the establishment of mitotic spindle, centrosome duplication, centrosome separation as well as maturation, chromosomal alignment, spindle assembly checkpoint, and cytokinesis. Required for initial activation of CDK1 at centrosomes. Phosphorylates numerous target proteins, including ARHGEF2, BORA, BRCA1 CDC25B, DLGP5, HDAC6, KIF2A, LATS2, NDEL1 PARD3, PPP1R2, PLK1 RASSF1 TACC3, p53/TP53 and TPX2. Reg µLates KIF2A tub µLin depolymerase activity. Required for normal axon formation. Plays a role in microtub µLe remodeling during neurite extension. Important for microtub µLe formation and/or stabilization. Also acts as a key reg µLatory component of the p53/TP53 pathway, and partic µLarly the checkpoint-response pathways critical for oncogenic transformation of cells, by phosphorylating and stabilizing p53/TP53. Phosphorylates its own inhibitors, the protein phosphatase type 1 (PP1) isoforms, to inhibit their activity. Necessary for proper cilia disassembly prior to mitosis.
